The "shake flask" method described is based on the OECD [Organisation for Economic Co-operation and Development] Test Guideline [i. e. the 1981 Test Guideline 107. ] This test method includes two separate procedures : the shake-flask method and high performance liquid chromatography(HPLC).... The initial solubilization of yolk platelets demonstrated that the lipid was present in the soluble fraction containing lipovitellin and phosvitin. These two proteins have been resolved and the lipovitellin fraction was shown to exhibit the absorption maximum at 375 nm that characterizes DCD [dorsalizing cytoplasmic determinant]. The lipovitellin was purified and its total lipids extracted and separated on HPLC [high-performance liquid chromatography]. A lipid was found that has identical elution properties from a C18 reverse HPLC column and mass as DCD. Therefore, lipovitellin, a zinc protein known to contain 17% lipid by weight, is the oocyte storage protein for DCD.

The sample is injected into a moving stream of solvent, which flows through a column and detector. The passage through the column separates the sample compounds by one of four different methods: adsorption, partition, ion exchange, or size exclusion. A special form of liquid chromatography, high-performance liquid chromatography(HPLC), has become especially important for the separation of complex mixtures of nonvolatile materials. Essentially, it involves the use of very small particle size in column packing, with high pressure to increase the rate of flow and shorten the separation time to a few minutes.

One of the most significant advances in chromatography in recent years has been the introduction of high performance liquid chromatography. On réalise ainsi des séparations beaucoup plus rapides avec une meilleure résolution. L'appareil est constitué d'une colonne en acier inoxydable d'une longueur de l'ordre de 200 mm et de diamètre intérieur de 4 mm, remplie de particules de taille inférieure à 20 mm; cette colonne est précédée d'une pompe à haute pression, permettant d'injecter le solvant sous une pression de plusieurs dizaines de bars. Des microvannes assurent l'introduction de quelques microlitres de solution à analyser, en tête de colonne.

To obtain the rapid separation of large quantities of materials, various techniques have been developed to produce continuous forms of chromatography. The easiest of these is the batch process of automatic sampling and fraction collecting applied regularly to GC [gas chromatography] and HPLC [high pressure liquid chromatography]. Other more truly continuous forms of chromatography have been developed for GC, including the use of circular columns, moving bed processes and radial flow columns.

A potent new approach which marries the inherent speed and resolving power of HPLC with the biological specificity of affinity chromatography.

The technique of HPLAC was used to resolve a mixture of horse liver alcohol dehydrogenase and pig heart lactate dehydrogenase by consecutive elution with appropriate ternary complexes.... The two enzymes were separated ... in near quantitative yield and in less than 5 minutes. Furthermore, the heart muscle (H4) and skeletal muscle (M4) isoenzymes of lactate dehydrogenase were also resolved with equal rapidity on the AMP-silica column by a gradient of NADH.

The general versatility of HPLAC has been amply demonstrated with the resolution of enzymes and isoenzymes, albumin, protein antigens and antibodies, carbohydrates, glycoproteins, nucleosides and nucleotides using microparticulate silica-immobilized nucleotides, boronates, immunoadsorbents, lectins and charge transfer ligands.
