Il s'agit d'empêcher les opsonines, ces protéines plasmatiques, de venir se fixer sur les nanovecteurs. Pour ce faire, l'équipe de Patrick Couvreur a fait appel au concept physico-chimique de la «répulsion stérique» qui consiste à greffer à la surface des nanoparticules des chaînes de polymères de polyéthylène glycol (PEG) hydrophiles et flexibles. Grâce à leur mobilité moléculaire, les chaînes de PEG repoussent - par le processus de répulsion stérique - les opsonines et les empêchent de se fixer à la surface des nanovecteurs, qui de ce fait, ne sont plus reconnus par le foie et la rate.
