Les hydrates sont ainsi composés d'un composé, le lisinopril dans le cas présent, et d'un certain nombre de molécules d'eau (2 ici); l'eau peut se trouver sous forme d'eau de cristallisation ou sous n'importe quelle forme. Le terme hydrate est une exception à la règle de nomenclature qui veut que la terminaison en «ate» désigne un anion : le terme hydrate désigne une molécule d'eau et non l'ion OH- qui est nommé «hydroxide». Dans un nom de composé d'addition, les molécules d'eau sont toujours citées en dernier lieu. Une molécule d'eau est signalée par le terme «monohydrate»; les préfixes multiplicatifs «di-», «tri-», «tétra-», etc., sont utilisés pour indiquer le nombre de molécules d'eau (2,3,4, etc.). Kinesin is a dimeric motor protein that can move for several micrometers along a microtubule without dissociating. The two kinesin motor domains are thought to move processively by operating in a hand-over-hand manner, although the mechanism of such cooperativity is unknown. Recently, a ~50-amino acid region adjacent to the globular motor domain(termed the neck) has been shown to be sufficient for conferring dimerization and processive movement. Based upon its amino acid sequence, the neck is proposed to dimerize through a coiled-coil interaction. To determine the accuracy of this prediction and to investigate the possible function of the neck region in motor activity, we have prepared a serie of synthetic peptides corresponding to different regions of the human kinesin neck and analyzed each peptide for its respective secondary content and stability. 2, record 3, English, - synthetic%20peptide

De nombreux peptides amphiphiles existent dans la nature. Sous forme d'hélice, ils interagissent avec les bicouches lipidiques constituant les membranes cellulaires. En s'appuyant sur ce phénomène biologique, nous avons synthétisé en phase solide un peptide amphiphile contenant 26 acides aminés. C'est un peptide simple, constitué uniquement de deux acides aminés différents. Une chaîne formée de 20 résidus d'alanine constitue la partie hydrophobe et 6 résidus de lysine assurent l'hydrophilicité du peptide. Un tel peptide devrait avoir un comportement comparable aux peptides d'origine naturelle et être capable de s'insérer dans une bicouche lipidique. Nous étudions donc la conformation adoptée par ce peptide par différentes méthodes spectroscopiques telles que le dichroïsme circulaire et l'infra-rouge dans différentes conditions. Nous essayons ainsi de déterminer le pourcentage d'hélices présents dans ce peptide. Because chymotrypsin is secreted into the small intestine, it was first thought to be specific for the hydrolysis of relatively long polypeptides formed by the action of pepsin on ingested proteins in the stomach, but later work revealed that chymotrypsin can also attack short peptides. As well, we are continuing to explore a new approach to subunit vaccine design introduced by our laboratory a number of years ago(see Carayanniotis and Barber(1987) Nature 327, 59). By conjugation of purified protein or synthetic peptide antigens to monoclonal antibodies specific for defined cell surface determinants in vivo(e. g. class II MHC), it is possible to induce an effective antibody response to the "targeted" antigen(for recent data see Skea and Barber(1993) J. Immunol. 151, 3557). We are currently using recombinant DNA approaches to "engineer" specific B-and T-cell epitopes into selected sites on immunotargeting antibodies. The carrier determinant in the N-and C-region of hen egg-white lysozyme(lysozyme), recognized by T cells, which helps antibody formation to a single DNP residue bound to the Lysine-33 of lysozyme, was analyzed. P17(sequence 1--27 : Cys 6-CYS 127 : 123--129) and P17t(sequence 1-Homoser 12; Cys 6-Cys 127 : 123--129), which are known to be immunodominant antigenic determinants could induce helper T cells. When P17t was reduced and alkylated, one of the resultant peptides, P17tN(sequence 1-Homoser 12) could still induce helper T cells, but P17tC(sequence 123-129) could not. A synthetic peptide SP1-14(sequence 1-A1a 6--14) also induced helper T cells. Thus the carrier determinant helping in antibody formation to the DNP residue at Lysine-33 may be localized in the region corresponding to the sequence 1--12 of lysozyme. 2, record 8, English, - carrier%20determinant
