Les mutations prolongeant la vie peuvent agir sur l'une ou l'autre forme de sénescence (réplicative ou métabolique). Mais la plupart du temps, c'est la sénescence métabolique qui est ralentie. En règle générale, ces mutations nuisent au bon fonctionnement des gènes qu'elles affectent, sans toutefois les rendre totalement inefficaces.
