A protein secreted by transformed cells that can stimulate growth of normal cells.

A number of transforming growth factors have been isolated from media conditioned by tumor cells. Two of these were purified on the basis of their synergistic actions in stimulating the growth of fibroblasts in soft agar, an assay often used as an index of cell "transformation." Since cancer cells grow in soft agar, the growth factors were first thought to be mitogens involved in tumorigenesis and were called tumor growth factors. However their main role appear to be in embryonic development and in tissue repair.

Selectines facilitate the initial adhesion of leukocytes on the surface of vascular endothelium. This initial adhesion is considered to be the first step in the development of the atherosclerotic plaque. Adhered leukocytes migrate into the subendothelial space and form the so-called "foam" cells at a later stage. Three different selectine molecule have been identified: L-selectine is expressed on the leucocyte membrane, E-selectine is expressed on the "activated" endothelial cell membrane and P-selectine is expressed on both platelet and "activated" endothelial cell surface. P-selectine seems to play a role in the development of acute coronary syndromes. Increased serum levels of P-selectine have been found in both acute myocardial infarction and unstable angina.

Activin, a member of the transforming growth factor - beta family of peptides, is implicated in the regulation of cell growth and differentiation in a variety of biological systems. We have sought to identify immediate early genes whose altered expression may provide a common nuclear event involved in activin-regulated phenotypic changes in many cell types. In both human K562 myelogenous leukemia and rat PC12 pheochromocytoma cells, activin treatment caused transient transcription-dependent and protein synthesis-independent increases of junB messenger RNA (mRNA) within 1 hour ... Recently, several mesoderm-inducing factors (MIFs) have been discovered. These cause isolated Xenopus animal caps to form mesodermal cell types such as muscle, instead of their normal fate of epidermis. The MIFs fall into two classes. One comprises members of the fibroblast growth factor (FGF) family, and the other members of the transforming growth factor type beta (TGF- beta) family. Of the latter group, the most potent is XTC-MIF, a protein produced by Xenopus XTC cells. Here we show that XTC-MIF is the homologue of mammalian activin A.

Because of the prominent noevascularization observed in the growth of brain tumors, we studies the occurrence of basic fibroblast growth factor (bFGF) a potent angiogenic factor in astrocytomas, the most aggressive of which often have marked vascular hyperplasia. Using immunohistochemical methods, we examined 21 examples of such tumors, 7 glioblastomas multiform, 7 anaplastic astrocytomas, and 7 low grade astrocytomas. Results suggest a possible role for bFGF in tumor growth and in angiogenesis in astrocytomes.

Le TGF a qui présente une forte homologie avec l'epidermal growth factor stimule l'angiogénèse in vivo et est mitogène pour les cellules endothéliales. Le TGF B entraîne in vivo, après injection sous-cutanée, une accumulation de macrophages, de fibroblastes et de collagène; et induit une néovascularisation. En revanche, in vitro, il inhibe la croissance des cellules endothéliales. Son activité angiogénique in vivo met donc probablement en jeu les macrophages.
