Recombinant DNA (rDNA) molecules are DNA sequences that result from the use of laboratory methods (molecular cloning) to bring together genetic material from multiple sources, creating sequences that would not otherwise be found in biological organisms. Recombinant DNA is possible because DNA molecules from all organisms share the same chemical structure; they differ only in the sequence of nucleotides within that identical overall structure. Therefore, when DNA from a foreign source is linked to host sequences that can drive DNA replication and then introduced into a host organism, the foreign DNA is replicated along with the host DNA. 2, record 4, English, - recombinant%20DNA

Recombinant soluble CD4 (rsCD4, rCD4): rCD4 is a soluble form of the CD4 receptors attached to the surface of T4 cells. rCd4 is produced by recombinant DNA biotechnology using mammalian and insect cell lines. Human CD4 is the receptor that serves as a docking site for HIV viruses on certain lymphocyte cells. La fixation du V. I. H. à la cellule CD4+. La fixation de la particule virale sur la cible cellulaire se fait par interaction de la gp120 et de la molécule CD4, ce qui découvre la glycioritéine transmembranaire gp41. Une des extrémités de la gp41 s’introduit alors dans la membrane cellulaire et cet accrochage engendre la fusion entre la membrane virale et la membrane cellulaire. Le mécanisme de la pénétration intracellulaire du V. I. H. reste encore discuté. L'enveloppe virale conditionne donc l'infectivité du V. I. H. puisqu'il s’attache spécifiquement à la cellule qu'il infecte par l'intermédiaire de ses glycoprotéines d’enveloppe. Pour certains, la molécule CD4 n’ interviendrait pas cependant comme un récepteur fonctionnel du V. I. H. Après la liaison spécifique de la gp120 à la molécule CD4 à la surface cellulaire, le virus pénétre dans la cellule. Nous cherchons à identifier et à caractériser des gènes de suppression tumorale impliqués dans la progression du cancer de la prostate. Nous avons utilisé la fusion de microcellules pour transférer une portion du chromosome 12(nommé del(12)(q13) dans une lignée cellulaire cancéreuse humaine(DU145). Les hybrides cellulaires obtenus ne sont plus tumorigéniques lorsqu'injectés chez la souris nue. Ceci suggère la présence de gènes de suppression tumorale sur ce chromosome. Les protéines traduites de ces gènes auraient donc un effet inhibiteur sur l'établissement de tumeurs prostatiques. Nous avons utilisé une méthode de criblage différentiel(DDRT-PCR) pour identifier les ADN complémentaires exprimés préférentiellement dans les hybrides non tumorigéniques qui contiennent del(12)(q13). Ces ADN complémentaires furent isolés, réamplifiés, clonés et séquencés. Des analyses de types Northern démontrent une hausse d’expression de certains ARNm dans les hybrides non tumorigéniques.
