La biomasse microbienne est récupérée, généralement par centrifugation, lavée afin d'éliminer le milieu de culture retenu entre les cellules et séchée à 105 degrés Celsius, en présence de sable de Fontainebleau, à poids constant. Cette technique peu sensible ne s'applique qu'aux suspensions assez denses et dans lesquelles les microorganismes sont les seules particules en suspension. De plus, il n'est pas possible de faire la distinction entre biomasse vivante et biomasse morte.
