Les diverses conformations que l'on peut obtenir par rotation autour d'une liaison ne possèdent pas toutes la même énergie. Certains arrangements sont plus stables que d'autres: ainsi la molécule passe-t-elle plus de temps dans sa conformation de niveau énergétique le plus bas. Si l'on excepte quelques cas rares, la différence d'énergie entre les isomères conformationnels, appelés rotamères ou conformères [voir cette autre fiche], est très faible, il n'est donc en général pas possible de les isoler.
