Method devised by Brown which is particularly useful when, owing to its chemical nature, insolubilization of an enzyme cannot be achieved by glutaraldehyde crosslinking alone. The Bartels PRIMA SystemTM Cytomegalovirus IgM Enzyme Immunoassay is a rapid, qualitative method for the detection of IgM Antibody to cytomegalovirus in human serum. Patient serum is diluted with specimen diluent that contains an absorbent to remove IgG. The absorbent prevents IgG/rheumatoid factor complexes from forming and prevents interfering competition from IgG during the test procedure. The treated serum is incubated with purified CMV antigens bound to EIA plate wells. If antibodies to CMV are present, they bind to the antigen and do not rinse off. Subsequently, when enzyme labeled anti-human IgM is added to the reaction site, it binds to the immobilized antibodies. After washing and the addition of a chromogenic substrate and stop solution, specimens containing antibodies to CMV produce a color endpoint reaction which is measured and compared to negative and positive controls, and may be measured spectrophotometrically. 1, fiche 3, Anglais, - chromogenic%20substrate

Immobilization can result in significant changes in the properties of enzymes in comparison with the properties of the soluble enzyme. It is likely that the properties of the immobilized enzyme are nearer to the in vivo properties since dilute soluble enzyme studies normally ignore the complex realities of the intracellular environment. Grâce à leur spécificité et leur rapidité d'action dans des conditions douces, les enzymes peuvent constituer des réactifs très précieux offrant d'énormes possibilités. Cependant, leur emploi en solution reste limité en raison de leur fragilité (dénaturation) et de la difficulté de récupération de l'enzyme pour une utilisation répétée. The effect of pH on the stability of immobilized enzymes has been studied.... The stability of enzymes immobilized using polycationic carriers was enhanced on the acid side.... These shifts toward the acid side may be explained as follows. When an enzyme is bound to a polycationic carrier, positive charge on the enzyme protein increases, and the pH of the immobilized enzyme region becomes more alkaline than that of the external solution. An immobilized enzyme system is the material which results from immobilization of an enzyme onto a support matrix. Magnetic support matrices and immobilized enzyme systems offer several advantages when compared to nonmagnetic systems. For example, separation of such materials from other nonmagnetic solids by use of a magnetic field can permit separations otherwise difficult or impossible to perform. Such a situation is represented by the case where two enzymes, with different deactivation times, are used concurrently on a substrate, with one immobilized enzyme system magnetic and the other nonmagnetic. A variety of enzymes have now been adsorbed to modified celluloses(DEAE) which, in two instances, have led to the development of major industrial-scale immobilized enzyme processes. In one case Tosa et al. brought to industrial scale a process in which various amino acylases adsorbed to DEAE cellulose or DEAE Sephadex are employed to convert tonnage quantities of synthetic DL amino acids to the biologically available L-form. Le terme cellules immobilisées couvre aussi bien les cellules non viables que celles ayant conservé la possibilité de pouvoir se diviser après l'immobilisation. Dans le premier cas (...), les microorganismes fragilisés par l'utilisation de réactifs chimiques deviennent des "sacs à enzymes" et sont utilisés pour une réaction de transformation unique. Par contre, dans le second cas (...), avec des cellules viables, il est possible de mettre au point une véritable alternative aux procédés de la fermentation en mettant en jeu des séquences réactionnelles utilisant des cofacteurs et des métabolites intermédiaires. The carrier-binding method is the oldest immobilization method for enzymes.... The amount of enzyme bound to the carrier and the activity after immobilization depend markedly on the nature of the carrier. Although the selection of a carrier also depends on the nature of the enzyme itself, the following aspects must be considered;(1) particle size,(2) surface area,(3) molar ratio of hydrophilic to hydrophobic groups, and(4) chemical composition. Where the enzyme is expensive then the extra cost of immobilization, with its attendant promise of catalyst re-use, may be less than the cost of continually adding more enzyme to the reaction system. Thus the criterion of re-use of enzyme is important; so too is the stability of the immobilized enzyme in relation to the average residence time of substrate within the reactor. In order to obtain a high yield of product a balance must be struck between flow rate, reactor volume, and enzymic activity. High flow rates and small reactor volumes will give a small residence time and thus, for high product yield, will require a highly active enzyme preparation. When the activity ratio of HK and G-6-PDH is small, the difference between immobilized and free enzymes becomes great..., while when it is high, the difference becomes small. In the latter case, it is considered that as the concentration of G-6-P within the carrier becomes very high due to the high activity of HK, the difference between the two systems becomes less apparent, because rather large amounts of G-6-P are released into the outer solution against the diffusion resistance.
